Determination of the amount of experimental animals administered

When observing the effects of a drug, the dose that should be given to the animal is an important issue that should be determined at the beginning of the experiment. The dose is too small, the effect is not obvious, the dose is too large, and it may cause animal poisoning to death. The dosage can be determined as follows:


1. The toxic dose or lethal dose is roughly explored with a mouse, and then the dose is less than the amount of poisoning, or a fraction of the lethal dose is used, which is generally 1/10-1/5.


2. The dosage of the crude botanical preparation is more than the conversion of the crude drug.


3. For chemistry, reference may be made to known drugs having similar chemical structures, particularly drugs having similar chemical structures and actions.


4. After the dose is determined, if the effect of the first experiment is not obvious, and the animal has no symptoms of poisoning (weight loss, lack of energy, decreased activity or other symptoms), the dose can be re-tested. If poisoning occurs and the effect is obvious, the dose should be lowered again. In general, the effect of a drug often increases with increasing dose in a suitable dosage range. Therefore, when conditions are met, it is best to use several doses at the same time for experiments to obtain more complete information on the effects of the drug. If there is no regularity between the dose and the intensity of the effect in the experimental results, it should be carefully analyzed.


5. When experimenting with large animals, the initial dose can be used in one-fifth to one-half of the dose of the mouse, and the dose can be adjusted later depending on the response of the animal.


6. When determining the dose to be administered to an animal, the age and physical strength of the animal to be administered should be considered. Generally, the determined dose to be administered refers to an adult animal, and if it is a young animal, the dose should be reduced. For example, in the case of dogs: when the dosage of dogs for 6 months or more is 1 part, 1/2 part for 3-6 months, 1/4 part for 45-89 days, and 1/8 for 20-44 days. For the 10-19 days, give 1/16 copies.


7. When determining the dose of the animal, it is necessary to consider the dosage according to the route of administration. When the oral dose is 100, the enema amount should be 100-200, the subcutaneous injection amount is 30-50, and the intramuscular injection amount is 25-30. The amount of intravenous injection is 25.
