Golvatinib (E7050) In vitro studies:

In vitro studies have shown that E7050 can effectively inhibit the phosphorylation of c-Met and VEGFR-2. E7050 also potently inhibits c-met-proliferating tumor cells as well as HGF- or VEGF-stimulated endothelial cell growth. All exogenous and endogenous HGF-induced reversible, irreversible, and mutation-selective EGFR-TKIs in lung cancer cell lines that act on EGFR mutations are avoided in vitro by E7050 by blocking the Met/Gab1/PI3K/Akt pathway. E7050 can also prevent the gefitinib resistance of HCC827 cells due to the continuous action of HGF.

Golvatinib (E7050) In vivo studies:

In vivo experiments confirmed that E7050 inhibited the phosphorylation of c-Met and VEGFR-2 in tumors and strongly inhibited tumor growth and tumor angiogenesis in xenograft models. Treatment of high-dose E7050 (50-200 mg/kg) tumor cell lines containing c-met expansion induced tumor recovery and disappearance. In the peritoneal model E7050 has anti-peritoneal tumor activity and can prolong the lifespan of treated mice. In another xenograft model study, Ma-1/HGF-induced tumors had more angiogenesis than control tumors and had resistance to gefitinib. E7050 alone can inhibit angiogenesis and tumor growth in Ma-1/HGF tumors. The combination of E7050 and gefitinib significantly induced tumor regression.
