Zebrafish is a model system widely used in research and drug discovery, and is popular for its similarity to human disease phenotypes. Many imaging-based experiments can be used to determine phenotypic changes in zebrafish, but manual acquisition and analysis of images is cumbersome and time consuming. Automation of these processes will greatly increase throughput and data quality. Here, we show how Operetta CLSTM and Opera PhenixTM perform high-throughput imaging and phenotypic analysis and screening of zebrafish, allowing you to focus on data evaluation rather than data generation.
Automatically detect high resolution images - easily lock zebrafish
The positioning and imaging of zebrafish in 96-well plates with a conventional microscope is very time consuming. Harmony® high-content analysis software combines PreciScanTM intelligent image acquisition tools, integrated low-magnification pre-scan workflow, image analysis, and higher magnification rescan, fully automated to reduce acquisition time and invalid data volume, and Significantly speed up the analysis. The pre-scanned positioning area is scanned with a 20x (1.0 NA water immersion lens). The Z-axis confocals the maximum projection of 50 layers. The acquisition time of 700 images is <2 minutes. Automatic detection and acquisition means unattended.

Automatically and accurately determine morphological features
Harmony ® imaging and analysis software makes it easy to detect changes in fluorescence intensity, morphology and texture. The example (Figure 4) shows that a single fluorescent dye can be used to identify different body parts. Identify the head and spine based on the width. The equivalent ellipse is used to describe the shape of the spine and estimate its curvature. The tail axis (Figure 5) automatically detects the tail curvature in a similar manner.
